Could you please tell us about your research?

It is research to detect interactions between proteins and various low-molecular compounds, including anticancer drugs. When low-molecular compounds are administered into a living body, they interact with various proteins involved in absorption, distribution, metabolism, excretion (ADME) to show the bioactivities, including main effects and side effects. The goal is to identify the proteins with which each compound interacts and to comprehensively clarify its behavior in the living body.

What kind of measurement are you doing by using AFFINIX series?

We use AFFINIX series as a base for screening peptides and proteins that interact with low-molecular compounds by using T7 phage display (PD) method.

The response to the binding of the compounds to the phage is very small, so immediate tracking of the interaction by QCM with high detection sensitivity is very useful.

Moreover, AFFINIX has overcome many technical problems of T7PD and has improved accuracy.

Could you tell me the features of AFFINIX series from your point of view?

Because of its simple structure and easy maintenance, it is very easy to use for students and beginners.

The cuvette-type structure is also ideal as a base for PD method and other display cloning technologies.

Other than that, by combining AFFINIX with other experimental techniques, AFFINIX can improve their performance, and AFFINIX can be used as an interaction measurement tool in a wide range of research. I think the possibilities of AFFINIX will be unlimited, depending on ideas.

Could you please tell us about your research?

Our research is the analyzation of the interaction mechanism between synthetic polymers and biopolymers.
Specifically, we synthesize synthetic polymers with different viscoelasticity and evaluate how viscoelasticity affects the binding and dissociation rates of proteins with synthetic polymers.

What kind of measurement are you doing by using AFFINIX series?

We use AFFINIX QN Pro to analyze the viscoelasticity of polymer nanoparticles that are synthesized. We also use AFFINIX Q4 to analyze the binding and dissociation rate constants of proteins to polymer nanoparticles.

Could you tell me the features of AFFINIX series from your point of view?

AFFINIX QN Pro is very useful because it can easily evaluate the viscoelasticity of nanometer-sized high polymer materials. Four experiments can be conducted simultaneously by using AFFINIX Q4, so control experiments and reproducibility tests can be performed in one day. The efficiency of the research has increased dramatically.

Could you please tell us about your research?

We develop modification methods of glass and metal surfaces using sugar chains and high polymers.

What kind of measurements are you doing by using AFFINIX series?

The surface of QCM sensor for AFFINIX Q4 that is coated with the glass and metal is modified with sugar chains and high polymers. The modification amount of the high polymer is quantified by AFFINIX Q4. AFFINIX Q4 is also used to quantify the interactions of lectins with high polymer-modified surfaces.

Could you tell me the features of AFFINIX series from your point of view?

The AFFINIX Q series offers sensor cells that are modified with glass, titanium, and aluminum. It is very convenient for surface modification research of inorganic materials. It is also very useful that AFFINIX Q determines the absolute value of the molecular mass attached to the surface of the substrate and can measure four samples at the same time.

Could you please tell us about your research?

It is a research of artificial nano-assemblies composed of proteins or protein-nucleic acids that are materials for biomaterials and biodevices.

What kind of measurements are you doing by using AFFINIX series?

It is used to quantitatively evaluate the accumulation of artificial assemblies of proteins or protein-nucleic acids fixed with thiol groups in the form of a gold electrode.

Could you tell me the features of AFFINIX series from your point of view?

I think AFFINIX is very easy to measure. Before purchasing AFFINIX, our group had never used QCM, but once we clear the first condition search, we get excellent data now.

Could you please describe the overview of your research ?

Our research is involved with developing methods for producing robust synthetic polymer nano particles with antibody-like affinity and selectivity for targeted peptides and proteins. We recently demonstrated that a synthetic polymer nanoparticle can function in vitro and in vivo to neutralize the deadly function of a toxic peptide. We are pursuing applications of these "plastic antibodies" in diagnostics, protein purification and as antibody replacements for therapeutic applications.

What kind of experiments are you doing by using AFFINIX Q4 ?

We use the AFFINIX Q4 for screening the affinity of our plastic antibody nanoparticles against target biomolecules. We also analyze the kinetics of binding with the AFFINIX Q4. These studies have resulted in the successful development of "plastic antibodies" against the toxic peptide melittin (major component of bee venom), mouse IgG, and an allergenic protein of Japanese cedar pollen allergy.

Could you please tell us features and benefits about AFFINIX system ?

Sensitivity, the AFFINIX Q4 operates at "therapeutic" concentrations. Other methods such as Isothermal Titration Calorimeter (ITC), require concentrated samples that consume large quantities of protein. In contrast, the AFFINIX Q4 needs only modest amounts of sample. The AFFINIX Q4 can run 4 measurements simultaneously for rapid and efficient sample screening.
